Role of exosomes from irradiated colorectal cancer cells on “bystander effect” mediated receptor cells.

Abstract

Introduction: One of the treatments for colorectal cancer is radiotherapy, which despite its many advantages, has significant adverse effects. One of these is the so-called Bystander effect, in which irradiated cells transmit signals to cells not exposed to radiation, which modify their function and are associated with the formation of new primary tumours in areas distant from the initial tumour. Recently, exosomes have been proposed as mediators of the Bystander effect, however, their contribution is not yet fully elucidated. Objective: In this study, the effect of exosomes released from colorectal cancer cells in response to 0,6 Gy irradiation was evaluated in cells derived from normal colon tissue. Methodology: Cell culture techniques were used; ultracentrifugation to obtain exosomes; quantification of cell death by typan blue; Rt-q-PCR for the expression of anti-apoptotic transcripts. Result: Exosomes from cancer cells induced cell death progressively over time and a downward trend of anti-apoptotic transcripts BCL-2 and BCL-W. Conclusion: We observed that exosomes released from irradiated colorectal cancer cells induce the death of non-irradiated normal colon cells progressively over time and a downward trend of anti-apoptotic BCL-2 and BCL-W transcripts, indicating a possible association with the apoptotic pathway.
